This protocol describe the optimisation of a commercial kit "DNeasy Plant Pro Kit, QIAGEN" reserved to gDNA extraction from plant. This protocol provides clear, straightforward, and transparent mandatory steps to ensure reproducibility and repeatability in the manual extraction of genomic DNA (gDNA) from mycelium liquid cultures or mushroom tissues. It has been successfully tested across various species belonging to the Basidiomycota and Ascomycota fungi phyla.
